Description

In this module, synthetic routes to a number of common active pharmaceutical ingredients will be examined as will the significance of drug impurity profiles associated with these routes. 

The fields of Medicinal Chemistry and Pharmacognosy, with related processes and technologies for drug discovery and design will be introduced and links between a drugs structure, its physicochemical properties and pharmacological activity will be highlighted. 

Learning Outcomes

On completion of this module the learner will/should be able to;

1.

Describe some of the principal routes used in the preparation of common pharmaceutical products and important intermediates.

2.

Explain the significance and consequence of impurities which may be encountered during pharmaceutical product synthesis and examine the characterisation and classification of such impurities.

3.

Relate a drug's chemical structure to its physicochemical properties and consequent entry, distribution, action and elimination from the body. 

4.
Interpret outcomes of simple QSAR studies and suggest suitable structural modifications to optimise potency.
5.

Demonstrate an understanding of the approaches, processes and technologies employed by the field of Medicinal Chemistry in drug discovery and design. 

6.

Discuss the significance of natural sources for drug discovery and the challenges associated with generating new development candidates from natural sources. 

Teaching and Learning Strategies

This module will be delivered full-time. This will include lectures and workshops augmented by independent learning and directed learning. This approach is expected to address student learning needs. Moodle will be used as a repository of educational resources and as a means of assessment (e.g. quizzes, uploading assignments and journals).

Module Assessment Strategies

This module is 40% continuous assessment and 60% final examination.

Continuous assessment will include a blend of essay or journal review and short answer questions.

Indicative Syllabus

Synthetic routes towards the synthesis of pharmaceutical products and intermediates - aliphatic- aromatic, substituted aromatic, heterocyclic compounds.

Pharmaceutical impurities including classification, characterisation and elimination. 

Physicochemical properties influencing pharmaceutic, pharmacokinetic and pharmacodynamic aspects of drug action.

Structural features and physicochemical properties of drug like molecules and druggable targets.

Rational drug design including the following elements SAR, QSAR, identification of pharmacophore,  structural analogues, strategies for lead optimisation.

The drug discovery and development process.

Drug discovery from natural sources including the following elements chemical diversity, existing drug classes from natural sources eg alkaloids bioprospecting, screening, isolation, characterisation.
